This paper demonstrates the validity of the Naess–Gadai method for extrapolating extreme value statistics of second-order Volterra series processes through application on a representative model of a deep water small size tension leg platform (TLP), with specific focus on wave sum frequency effects affecting restrained modes: heave, roll and pitch. The wave loading was estimated from a second order diffraction code WAMIT, and the stochastic TLP structural response in a random sea state was calculated exactly using Volterra series representation of the TLP corner vertical displacement, chosen as a response process. Although the wave loading was assumed to be a second order (non-linear) process, the dynamic system was modelled as a linear damped mass-spring system. Next, the mean up-crossing rate based extrapolation method (Naess–Gaidai method) was applied to calculate response levels at low probability levels. Since exact solution was available via Volterra series representation, both predictions were compared in this study, namely the exact Volterra and the approximate one. The latter gave a consistent way to estimate efficiency and accuracy of Naess–Gaidai extrapolation method. Therefore the main goal of this study was to validate Naess–Gaidai extrapolation method by available analytical-based exact solution. Moreover, this paper highlights limitations of mean up-crossing rate based extrapolation methods for the case of narrow band effects, such as clustering, typically included in the springing type of response.publishedVersio

Pyrene-based aggregation-induced emission luminogens and their applications

"Unity is force"-Aesop. It is a common phenomenon that traditional chromophores exhibit high fluorescence in dilute solutions, yet luminescence is quenched at high concentrations or in the aggregate state, i.e. "aggregation-caused quenching" (ACQ). Tang reported the unusual photophysical observation that luminogens can exhibit weak or no fluorescence in solution, yet they are highly emissive in the aggregate or solid state; this is defined as aggregation-induced emission (AIE). The discovery of AIE helped solve the ACQ effect in traditional luminophores. Pyrene is an important polycyclic aromatic hydrocarbon (PAH), which exhibits very different photophysical behavior in solution versus the aggregate state, and the ACQ effect has played a dominant role in pyrene chemistry. The ACQ effect is harmful for some practical applications and is a challenge in organic light-emitting diodes (OLEDs) and light-emitting electrochemical cells, for which the effect is more severe in the solid state. Thus, how to overcome the ACQ effect observed in pyrene chemistry still remains a challenge. In this review, we discuss how following basic AIE mechanisms such as the restriction of intramolecular motion (RIM), excited-state intramolecular proton transfer (ESIPT), and twisted intramolecular charge transfer (TICT), can transform pyrene-based ACQ luminogens to AIE luminogens with excellent optical properties. Furthermore, prospective applications of pyrene-based AIEgens are discussed, as is the potential for designing new organic functional materials

Objectives: Fallopian tube (FT) injury is an important factor that can lead to tubal infertility. Stem-cell-based therapy shows great potential for the treatment of injured fallopian tube. However, little research has shown that mesenchymal stem cells (MSCs) can be used to treat fallopian tube damage by in situ injection. In this study, we in situ transplanted PF127 hydrogel encapsulating dental pulp stem cells (DPSCs) into the injured sites to promote the repair and regeneration of fallopian tube injury.Materials and methods: The properties of dental pulp stem cells were evaluated by flow cytometry, immunofluorescence analysis, and multi-differentiation detection. The immunomodulatory and angiogenic characteristics of dental pulp stem cells were analyzed on the basis of the detection of inflammatory factor expression and the formation of capillary-like structures, respectively. The biocompatibility of PF127 hydrogel was evaluated by using Live/Dead and CCK-8 assays. The effects of PF127 hydrogel containing dental pulp stem cells on the repair and regeneration of fallopian tube injury were evaluated by histological analysis [e.g., hematoxylin and eosin (H&E) and Masson’s trichrome staining, TUNEL staining, immunofluorescence staining, and immunohistochemistry], Enzyme-linked immunosorbent assay (ELISA), and RT-PCR detections. Results: Dental pulp stem cells had MSC-like characteristics and great immunomodulatory and angiogenic properties. PF127 hydrogel had a thermosensitive feature and great cytocompatibility with dental pulp stem cells. In addition, our results indicated that PF127 hydrogel containing dental pulp stem cells could promote the repair and regeneration of fallopian tube damage by inhibiting cell apoptosis, stimulating the secretion of angiogenic factors, promoting cell proliferation, modulating the secretion of inflammatory factors, and restoring the secretion of epithelial cells. Conclusion: In this study, our results reported that in situ injection of PF127 hydrogel encapsulating dental pulp stem cells into the injured sites could provide an attractive strategy for the future treatment of fallopian tube injury in clinical settings

Intestinal autochthonous bacteria play important roles in the maintenance of the physiological homeostasis of animals, especially contributing to the host immune system. In the present study, the variation of autochthonous bacterial community in the intestinal tract of 2-7 months-old tiger pufferfish Takifugu rubripes and bacterial communities in the seawater of recirculating aquaculture system (RAS) and the following offshore sea cage aquaculture system (OSCS) were analyzed during the aquaculture period from May to October 2021. Proteobacteria was found to be the most dominant phyla in both intestinal and seawater bacterial communities, which accounted for 68.82% and 65.65% of the total bacterial abundance, respectively. Arcobacter was the most core bacterial taxon in the intestinal bacterial community, with the most dominant abundance (42.89%) at the genus level and dominant positions in co-occurrence relationships with other bacterial taxa (node-betweenness value of 150). Enterococcaceae was specifically enriched in the intestinal bacterial community of pufferfishes from RAS, while Vibrionaceae was enriched in the intestinal bacterial community from OSCS. The F-values of beta diversity analysis between intestinal and seawater bacterial communities generally increased from May (6.69) to October (32.32), indicating the increasing differences between the intestinal and seawater bacterial communities along with the aquaculture process. Four bacterial taxa of Weissella sp., Akkermansia muciniphila, Dietzia sp. and Psychrobacter pacificensis had significant correlations with immune response parameters, and they were suggested to be the indicators for immune status and pathological process of pufferfish. The knowledge about the specific core bacteria, potentially pathogenic bacteria and the change of bacterial community in the intestinal tract of cultured pufferfish is of great scientific significance and will contribute to the understanding of intestinal bacterial homeostasis and biosecurity practice in pufferfish aquaculture

Design of asymmetric flexible micro-gripper mechanism based on flexure hinges

Symmetric micro-gripper mechanism is very easy way to destroy the micro-components or parts for nonuniform force in the processing of micro-assembly. Aiming at the requirements of micro-assembly for the microtubule (diameter of 0–200 μm) components, a new type of asymmetric flexible micro-gripper mechanism based on flexure hinges was designed and studied. The asymmetric micro-gripper mechanism was driven by piezoelectric actuator, whose output displacement was amplified and transmitted by flexure hinges, and the flexible parallel four-bar mechanism was used to ensure that the micro-clamp moves and is held parallel. The displacement amplification ratio of the asymmetric flexible micro-gripper mechanism was deduced theoretically, and the finite element analysis and the experiment were also carried out to study the displacement amplification ratio in detail. The experiment results show that the displacement amplification ratio of the asymmetric flexible micro-gripper mechanism is 4.16, compared with the finite element analysis result and the theoretical calculation result; the error between them is 1.89% and 5.67%, respectively. The experiment results also show that the step-wise resolution of the micro-gripper is 7.50 μm, and the output force of the right micro-clamp was measured at 0.28 N. The asymmetric flexible micro-gripper mechanism is able to perform the micro-assembly tasks for the microtubule parts, and it is helpful to design this type of micro-gripper mechanism

Analysis of the Transaction Behavior of Live Broadcasters with Goods Based on the Multi-Stage Game under Dynamic Credit Index

    No full text
In the live streaming transactions, the subject’s trading breach of trust frequently appears, which affects the healthy development of the industry. Therefore, from the perspective of internal supervision and governance, aiming at the interest relationship, behavior strategy, and game relationship between the live platform and the anchor and brand, this paper constructs a multi-stage honest transaction game model of the three behavior strategies, establishes a dynamic credit index mechanism, proposes a complaint compensation and cost-sharing strategy for breach of trust based on the change in dynamic credit index, and explores the influence of relevant parameters on the trading strategies of subjects. The research found that the internal penalty factor and the weight of dishonesty cost sharing can effectively restrain the behavior of transaction subjects; and the increase factor of dishonest transaction income is an important factor affecting the choice of behavior strategies of transaction subjects. Coefficients, internal and external penalty coefficients, as well as implementing a governance strategy of parallel rewards and punishments for trading entities, will assist in regulating the behavior of trading subjects
